With the ChargeSwitch® EasyPlex™ gDNA Kit’s simple protocol, you’ll save time by going directly from genomic DNA (gDNA) isolation to PCR in one well, in less than 45 minutes.


High-Performance gDNA Without Elution, Contamination, and False Positives

ChargeSwitch® kits come with aqueous buffers that do not contain guanidine or ethanol, which can inhibit downstream applications. Genomic DNA can be isolated from as little as 10 µL whole blood or 1–2 x 104 cells per well. Purified gDNA is amplified directly from microplate wells or 8-well strips, without elution, and is suitable for downstream procedures such as multiplex PCR, qPCR, STR (short tandem repeat) analysis, WGA (whole genome amplification), and sequencing. In addition, ChargeSwitch® EasyPlex™ gDNA Kits are compatible with most automated liquid handling workstations, and contain semi-skirted microplates and 8-well strips that fit into most thermal- and real-time- cyclers for high-throughput procedures (Figures 1 and 2). With the ChargeSwitch® EasyPlex™ gDNA Kit, you get successful, highly-pure gDNA results using less sample, contamination, sample error, and false positives.

. Using the ChargeSwitch® EasyPlex™ gDNA Kit, you can lyse and purify gDNA from up to 96 samples in <45 minutes. Add PCR components directly to the wells of the same plate. The elimination of transfer steps means a reduced risk of contamination and sample mix-ups.

Genomic DNA was isolated from frozen blood using ChargeSwitch® beads (control) or the ChargeSwitch® EasyPlex™ gDNA Kit. An optimized primer mix for the CFTR gene (Tm Bioscience) and Platinum® Taq Polymerase were used to perform multiplex PCR directly in the wells of the coated plate. The amplified products were separated on a 4–12% gradient polyacrylamide gel, and stained with SYBR® Gold Nucleic Acid Gel Stain. Reactions performed in the wells of the ChargeSwitch® EasyPlex™ plate were of the same quality as reactions performed on ChargeSwitch® bead–purified template. ChargeSwitch® EasyPlex™ Automation Flexibility

The ChargeSwitch® EasyPlex™ gDNA Kit is easy to automate on most automated liquid handling work stations (Figure 3), without the need for any special equipment for processing (such as a vacuum manifold, shaker, or centrifuge).  Depending on your deck and pipetting configuration, a single plate can take as little as 30 minutes to process, after sample lysis.
