Feeder-dependent
Culturing PSCs on a layer of feeder cells in KnockOut™ Serum Replacement supplemented medium
A feeder layer, traditionally comprised of inactivated mouse embryonic fibroblasts (MEF), serves as an adherent surface for the PSCs. While the feeder layer does not replicate in the culture dish, it plays a supporting role in supplying required nutrients and matrix proteins for the PSCs.

Conditioned Medium
A KnockOut™ Serum Replacement medium that supports the growth of PSCs in the absence of a feeder layer.
Conditioned medium is KnockOut™ Serum Replacement supplemented medium that has been conditioned on a layer of feeder cells, and therefore can be used to grow PSCs in the absence of a feeder layer. In the absence of a feeder layer, an extracellular matrix is required, such as Geltrex® LDEV-Free hESC-qualified Matrix, in order for the PSCs to adhere to the growing surface.

Feeder-free
Nutrient-rich media that does not require a feeder-layer of cells
Feeder free media are nutrient-rich and do not require a feeder-layer of cells. KnockOut™ Serum Replacement supplemented medium is not recommended for growing PSCs feeder-free, however we do recommend the use of StemPro® hESC SFM.
In a feeder-free system, an extracellular matrix is required, such as Geltrex® LDEV-Free hESC-qualified Matrix.
Feeder-based, xeno-free medium:
Culturing PSCs in xeno-free conditions with feeders requires the use of human feeders, typically human foreskin fibroblasts.

Xeno-free
PSC culture media comprised of only human-origin or recombinant components
The removal of animal-derived components from PSC culture media provides a xeno-free medium which can facilitate the transition of PSC research to clinical research applications. The Gibco® Cell Therapy Systems (CTS™) brand offers a broad array of products for use in cell therapy applications, including media, reagents, growth factors, enzymes, cell selection/expansion reagents, and devices.
