Chemically modified hot-start AmpliTaq Gold® 360 DNA Polymerase has been optimized to amplify a wide range of targets while delivering the high specificity, yield, and reliability you have come to expect.
Platinum® Taq DNA Polymerase uses antibody-mediated hot-start to enable increased specificity, yield, and sensitivity with the fastest activation times. Although there are several types of hot-start enzymes, the most reliable and widely used techniques involve chemically modified hot-start or antibody-mediated hot-start enzymes.
How to Choose Between Chemically Modified & Antibody-mediated Hot-start Reagents
| Chemically modified hot-start reagents are best when performing high-throughput applications. These enzymes withstand longer periods at room temperature. However, the activation time is longer and the read lengths are shorter. | Antibody-mediated hot-start products deliver fast activation times and minimal template degradation, which promote the amplification of longer fragments. However, antibody-mediated chemistries tend to exhibit reduced specificity and involve longer setup times. |

Platinum® Taq DNA Polymerase High Fidelity
- Greater than six times higher fidelity than Taq DNA polymerase
- Amplification of fragments up to 20 kb

The accuracy of Platinum® Taq DNA Polymerase High Fidelity is provided by a mixture of Platinum® Taq DNA Polymerase and the proofreading (3´ > 5´ exonuclease activity) enzyme from Pyrococcus species, GB-D polymerase. PCR specificity is improved with the incorporation of Platinum® automatic “hot-start” technology. Platinum® Taq DNA Polymerase High Fidelity is especially effective for high-fidelity applications such as cloning or mutagenesis. Platinum® Taq DNA Polymerase High Fidelity predominantly produces PCR products with 3’-A overhangs for compatibility with TOPO® TA Cloning® Kits.
