New Click-iT® Lipid Peroxidation Imaging Kit
The Click-iT® Lipid Peroxidation Imaging Kit and stand-alone Click-iT® LAA reagent employ click chemistry and an alkyne-modified linoleic acid (linoleamide alkyne or LAA) to detect lipid peroxidation–derived protein modifications by fluorescence microscopy or high-content screening. When incubated with cells, Click-iT® LAA incorporates into cell membranes. Upon lipid peroxidation, LAA is oxidized and produces 9- and 13-hydroperoxy-octadecadienoic acid (HPODE). These hydroperoxides decompose to α,β-unsaturated aldehydes, which readily modify proteins at nucleophilic side chains, producing alkyne-containing proteins that can then be detected with fluorescent or biotinylated azides using click chemistry.
The Click-iT® Lipid Peroxidation Imaging Kit provides the tools needed to visualize the lipid peroxidation modifications using green-fluorescent Alexa Fluor® 488 azide. If you’d like to customize the click reaction, we also offer the stand-alone LAA reagent, as well as a variety of azide-modified detection reagents and the Click-iT® Cell Reaction Buffer Kit.

|Lipid peroxidation detection in hemin-treated cells. After incubation with 50 µM LAA in complete growth medium at 37°C for 30 min, bovine pulmonary artery endothelial (BPAE) cells were treated with 10 μM hemin to induce lipid peroxidation–derived protein modifications (right), or left untreated (left) for 2 hr at 37°C, then fixed and permeabilized. The click reaction was performed with 5 µM Alexa Fluor® 488 azide from the Click-iT® Lipid Peroxidation Kit for 30 min. Cells were stained with NucBlue® Live Cell Stain and imaged on a Zeiss® Axiovert inverted microscope using a 40x objective.|

CellLight® reagents combine the utility and selectivity of fluorescent proteins (CFP, GFP, and TagRFP) with the transduction efficiency of BacMam 2.0, enabling unambiguous staining of organelles and cellular structures in live mammalian cells. Premo™ FUCCI Cell Cycle Sensor is a fluorescent two-color sensor of cell cycle progression and division in live cells. Cells change from red in the G1 phase, to yellow in G1/S interphase, to green in the S, G2, and M phases. Both CellLight® and Premo™ FUCCI reagents are easy to use—simply add, incubate, and image.

SYBR® GreenER™ Dye Shines in Real-Time PCR Master Mix
SYBR® Select Master Mix is a superior, cost-effective solution for your real-time PCR applications, delivering highly specific and sensitive quantitation with reliable single-copy detection. SYBR® Select Master Mix offers significant improvements over other master mixes by employing AmpliTaq® DNA Polymerase UP (Ultra Pure), which is activated by a proprietary hot-start mechanism. SYBR® Select Master Mix is also formulated with SYBR® GreenER™ dye—which is less inhibitory to PCR than SYBR® Green I dye, resulting in brighter signals—as well as heat-labile uracil-DNA glycosylase (UDG) for worry-free carryover contamination control. We offer SYBR® Select Master Mix in six sizes, from a 1 mL tube (100 reactions) to a 50 mL bottle (5,000 reactions).

Melt curves for 8 different amplification reactions (4 replicates each) using the SYBR® Select Master Mix. Single Tm peaks were obtained for all assays, regardless of amplicon length and GC content, demonstrating that all assays generated a single amplicon without primer-dimer or nonspecific amplification.
New Murine CD Antibody Markers for Flow Cytometry
Life Technologies now offers you several choices of fluorescent CD45 allele–specific monoclonal antibodies, providing more options for your multiplex flow cytometry experiments. The protein tyrosine phosphatase CD45 is an essential regulator of leukocyte activation and development. CD45 exists in allelic forms CD45.1/Ly-5.1 and CD45.2/Ly-5.2. Our new CD45 allele–specific antibodies can provide critical information when the resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation.

CD45.2 detection in BALB/c splenocytes by flow cytometry. Splenocytes were stained with 0.5 μg Mouse IgG2a,κ Isotype Control PerCP-Cy®5.5 (open histogram) or 0.5 μg Anti–Mouse CD45.2 PerCP-Cy®5.5 (filled histogram). Cells in the lymphocyte gate were used for this analysis.
