Invitrogen™ Luminex® Assays for the Met Signaling Pathway
To facilitate the analysis of STAT3 as well as a broad range of important signaling molecules, Life Technologies has developed a series of singleplex and multiplex immunoassays for the Luminex® platform. Samples containing targeted analytes bind to antibodies immobilized on color-coded polystyrene or magnetic beads. The beads are then analyzed with the Luminex® 100™ or 200™ system or the FLEXMAP 3D® system. By monitoring the spectral properties of the beads and the amount of associated RPE fluorescence (from the secondary antibody conjugate), | the concentrations of individual analyte targets in both singleplex and multiplex assays can be determined simultaneously. This workflow helps save cost, time, and sample. A member of the tyrosine kinase superfamily, c-Met is the receptor for hepatocyte growth factor, also known as scatter factor (HGF/SF). Phosphorylation of tyrosines 1234 and 1235 of c-Met is required for activation of the receptor’s tyrosine kinase activity. Several signaling proteins are phosphorylated and/or localized in response to c-Met phosphorylation. STAT3 (signal transducer and activator of transcription 3) is phosphorylated on tyrosine residue 705. STAT3 mediates the expression of a variety of genes in response to cell stimuli, and thus plays a key role in many cellular processes such as cell growth and apoptosis.

Interrogate c-Met Proteins for Cancer Research Using ELISA
c-Met ELISA kits allow easy and quantifiable measurement of soluble c-Met or phosphorylated c-Met. c-Met is a receptor tyrosine kinase for hepatocyte growth factor (HGF). Soluble c-Met is a truncated form of the c-Met membrane receptor and can be measured in serum, plasma, or supernatant. Phosphorylated and total c-Met can be measured in cell lysate samples. Receptor tyrosine kinases are important therapeutic inhibition targets for cancer research.
| c-Met has been shown to be overexpressed in a number of malignancies. Ligand binding of HGF to c-Met receptor triggers phosphorylation of tyrosines 1234 and 1235, leading to a cascade of downstream events including proliferation, angiogenesis, migration, and metastasis. The c-Met ELISA kits are valuable assay tools for studying the effects of drug inhibitors on c-Met signaling. For normalizing c-Met content of the samples, the c-Met (Total) ELISA Kit is also available, which assays for c-Met independent of phosphorylation status. The assay is completed using a 4-hour incubation protocol.
