The iBlot® Western Detection Kits work with the iBlot® Dry Blotting System, enabling complete western detection in less than 25 minutes. The kits are available with anti-mouse or anti-rabbit secondary antibodies and are compatible with chemiluminescent and chromogenic detection.

What Are the Components of the Kits?

The iBlot® Western Detection Kits consist of iBlot® detection stacks and iBlot® detection reagents. The reagents include all solutions needed to perform western detection—antibody diluent, wash solution, secondary antibody conjugated with alkaline phosphatase, and detection substrate (chemiluminescent or chromogenic). The top (cathode) and bottom (anode) stacks are provided as well as all other supplies required for the iBlot® western detection protocol. The only user-supplied reagent is the primary antibody.

What Is the Performance of the Kits?

The iBlot® Western Detection Kits offer comparable or better sensitivity than the conventional protocols for the majority of the antibody-antigen pairs while providing significant time savings. They enable complete western detection in less than 25 minutes, rather than 3–5 hours. For some antibody-antigen pairs optimization of the detection conditions is required. Figure 1. Comparison of iBlot® Western Detection Kit to WesternBreeze® Kit. Proteins from an SW480 (human colon adenocarcinoma cell line) lysate were transferred using the iBlot® Dry Blotting System. Mouse anti-p53 antibody was used as the primary antibody. Detection was performed with (A) the WesternBreeze® Chemiluminescent Kit—Anti-Mouse or (B) the iBlot® Western Detection, Chemiluminescent Kit (Anti-Mouse). The iBlot® Western Detection kit detected the proteins with sensitivity comparable to the WesternBreeze® kit.
