Overview of ProtoArray® Human Protein Microarrays
![]() | The ProtoArray® Human Protein Microarray is an advanced, high-content, functional protein microarray that enables rapid profiling of thousands of biochemical interactions in as little as one day. Researchers can use the ProtoArray® microarray to perform a variety of assays including biomarker identification, drug target discovery, enzyme substrate identification, antibody specificity profiling, and protein-protein interaction studies. Absolute Identification of Your Protein Targets with ProtoArray® Human Protein Microarray
Each ProtoArray® protein array has over 9,000 human proteins printed in duplicate on a one inch by three inch slide. The proteins are produced from clones selected from the Invitrogen Ultimate™ ORF Collection, the first and only clone resource to offer full-insert sequenced clones in a Gateway® vector. Proteins are expressed as N-terminal GST fusion proteins using a baculovirus expression system. After protein expression, proteins are purified under nondenaturing conditions. The proteins are printed at +4°C on an ultrathin layer of nitrocellulose to preserve native protein structure without the need for chemical fixation. Each slide is labeled with a unique bar code for easy tracking.
Individual microarrays on nitrocellulose-coated glass slides allow for the greatest flexibility in experimental design, with protocols for each supported application available. Some of these profiling applications (Figure 2) include:- Immune Response Biomarker Profiling — Biomarker Discovery for rapid discovery of disease markers specific to a variety of infectious diseases, cancers, and autoimmune diseases
- Enzyme Substrate Profiling — Profiling of kinases, ubiquitin ligases, and methyltransferases for target discovery and validation
- Small Molecule Profiling — Profiling of radiolabeled, biotinylated, or fluorescently labeled small molecules for target identification or selectivity
- Protein-Protein Interaction Profiling — Protein-Protein Interactions for mapping biochemical pathways
- Antibody Specificity Profiling — Antibody specificity profiling for research and therapeutic antibody development

Profiling With ProtoArray® Protein Microarrays
Using the ProtoArray® Human Protein Microarray is simple — and you get high-quality results in a fraction of the time. Regardless of your study, fluorescent applications can be completed in as little as one day. ProtoArray® microarray applications follow the same simple workflow of incubating a sample on the array, probing with a detection reagent, and then visualizing with a fluorescent scanner or phosphorimager (Figure 3).
