Real-Time PCR Master Mix Selection Guide

TaqMan® Master Mixes

TaqMan® Master Mixes contain the buffers, dNTPs, internal reference dye, and - most importantly - hot-start DNA polymerase, that power real-time PCR. They were developed for use with our comprehensive selection of TaqMan® Assays to streamline reaction setup and therefore minimize variability among replicates, assays, and experiments.

SYBR® Green Master Mixes

In SYBR® Green-based real-time PCR, amplification products are detected with fluorescent signal released by SYBR® Green dye, which fluoresces only when bound to double-stranded DNA. Our SYBR® Green Master mixes include buffers, dNTPs, internal reference dye, hot-start DNA Polymerase, and of course, SYBR® Green dye.

Reverse Transcription for Real-Time PCR

RNA must be converted to cDNA in order to be amplified using PCR. We recommend the Invitrogen® SuperScript® VILO™ cDNA Synthesis Kit for reverse transcription prior to analyzing gene expression (mRNA), long noncoding RNA, and pri-miRNA using real-time PCR. For analysis of microRNA and other small RNAs using TaqMan® Assays, we recommend the TaqMan® MicroRNA Reverse Transcription Kit.

One-Step Real-Time RT-PCR Kits

In one-step RT-PCR, reagents and enzymes for both reverse transcription (RT) and real-time PCR are added to RNA samples at the same time. Reactions are first incubated using conditions favorable for RT, then the resulting cDNA is immediately run in a real-time PCR reaction.

Real-Time PCR Directly from Samples

With Sample-to-Answer Kits, crude sample lysates can be used directly in real-time PCR or one-step RT-PCR. No nucleic acid purification is required, saving time and reducing the chance of sample mixup or cross-contamination.  We recommend the Ambion® Cells-to-CT™ Kits for gene expression and miRNA studies.  For SNP genotyping we recommend the TaqMan® Sample-to-SNP™ Kit.

High Resolution Melt (HRM) Master Mix

Mixed with your DNA sample and PCR primers, MeltDoctor™ HRM Master Mix provides the reagents needed to discriminate subtle differences in the target DNA sequence using post-PCR melting analysis.
