Platinum® Pfx DNA Polymerase
Platinum® Pfx DNA Polymerase is a proprietary enzyme preparation containing recombinant DNA polymerase from Pyrococcus sp. Platinum® Pfx DNA Polymerase possesses a proofreading 3'-5' exonuclease activity and provides higher fidelity than that of Pfu or PfuTurbo® DNA polymerases.
Platinum® Pfx DNA Polymerase Technology
Platinum® Pfx DNA Polymerase is a highly processive enzyme and possesses a fast chain extension capability. Due to specific binding of the antibody, Platinum® Pfx DNA Polymerase is provided in an inactive form. This reagent provides an automatic "hot start" for PCR. It increases specificity, sensitivity, and yield while allowing room temperature reaction setup. The antibody-bound enzyme is sufficient to eliminate or reduce non-specific amplification in PCR. Although polymerase activity is blocked at ambient temperatures, it is regained after the denaturation step in PCR cycling at 94°C. The high accuracy, specificity, and yield offered by Platinum® Pfx DNA Polymerase is ideally suited for demanding PCR applications such as site-directed mutagenesis and PCR expression cloning.

|Human genomic DNA (0 to 100 ng) was amplified with 0.3 μM each primer (for thrombospondin), 0.3 mM dNTPs, 1X Pfx Buffer, 1mM MgSO4, and 1.25 units Platinum® Pfx DNA Polymerase for 35 cycles.|
Better Fidelity and Yield Over a Wide Range of Target Size
|A sample of 200 ng K562 genomic DNA was amplified in duplicate with 1.25 units of Platinum® Pfx DNA Polymerase of 2.5 units of PfuTurbo DNA Polymerase, Platinum® Pfx DNA Polymerase reactions were assembled at room temperature; PfuTurbo reactions were assembled on ice. Targets were a 256 bp human beta globin (HBG); 1.0 kb, 1.8 kb, 5.2 kb, and 8.4 kb human myosin heavy chain (HMY HC); 2.7kb, 4.5 kb, and 7.5 kb myelin ologodendrocyte glycoprotein (MOG); and 802 bp thrombospondin (TS) heavy chain gene.|
High yield Without the Extra Bands
|A sample of 200 ng K562 genomic DNA was amplified in duplicate with 1.25 units of Platinum® Pfx DNA Polymerase or 2.5 units of PfuTurbo DNA Polymerase for 35 cycles. Platinum® Pfx DNA Polymerase reactions were assembled at room temperature; PfuTurbo reactions were assembled on ice. Targets were a 3.1-kb fragment of phenol sulfonyl transferase and a 5.2-kb fragment of myosin heavy chain.|
