There are two approaches for RT-PCR: two-step RT-PCR and one-step RT-PCR. Two-step RT-PCR is useful for detecting multiple messages from a single RNA sample. One-step RT-PCR is easier to when processing large numbers of samples and helps minimize carry-over contamination because tubes are not opened between cDNA synthesis and amplification.
