DRB
Strategy 1:
The Secore® DRB1 Locus Sequencing Kit (single amplification system) begins with only one PCR per sample. This method eliminates the need for confirmatory agarose gel and offers the user the most high throughput route for DRB1 sequencing.
Strategy 2:
The SeCore® DRB1, DRB3, 4 and 5 Sequencing Kit (Group Specific Set) provides eleven group specific amplification mixes. All eleven amplification mixes can be used for each sample, or the user can select only the amplification mixes necessary for each sample based on previous low resolution data. The resulting typing data for each separated allele is a greatly simplified homozygous electropherogram, eliminating the challenge of interpreting polymorphic positions. This strategy reduces the cis/trans ambiguities for DRB locus by 98%.
Additional Resolution:
DRB1 ambiguities can be further resolved by SeCore® Group Specific Sequencing Products (GSSP). The amplicon from either SeCore® DRB core kits serves as the template. GSS-Products contain a single group specific unidirectional sequencing mix and precipitation buffer. Sequencing parameters are identical to the other core SeCore® products.
DPB1 and DQB1
The DQB1 consists of separate amplifications of exon 2 and exon 3 followed by 4 sequencing reactions to consistently resolve all alleles.
