GIBCO® serum-free medium and Wave disposable bioreactors deliver superior insect cell growth and robust protein expression compared to a glass-lined bioreactor.
(Panel A) Growth and viability of Sf9 cells grown in Sf-900™ III medium in Wave Cellbag® bioreactor chambers and CelliGen Plus® bioreactors. Preadapted Sf9 cells were used to inoculate a Wave Cellbag® Bioreactor Chamber and a CelliGen Plus® Bioreactor at a seeding density of 4 × 105 cells/ml. Cell number and viability were monitored daily for 8 days. Each data point represents a single sample.
(Panel B) Sf9 cells were grown under conditions identical to those described in A, with introduction of a recombinant baculovirus containing the lacZ gene at day 2 post-inoculation (Sf9 density ~2 x 106 cells/ml, baculovirus MOI ~5). Cell number and viability were monitored daily for 8 days. Each data point represents a single sample.
(Panel C) Cell samples (0.5 ml) were taken daily for 4 days post-infection from the Wave Cellbag® bioreactor chamber and a CelliGen Plus® bioreactor described in Figure 2B and assayed for ß-galactosidase activity.
