With the ChargeSwitch® EasyPlex™ system, the entire sample is concentrated on the plate surface, enhancing sensitivity by eliminating dilutions of small amounts of nucleic acid. Without dilution, purity of bound nucleic acids is paramount in order to avoid a negative impact on detection in real-time PCR. To demonstrate the purity and sensitivity of bound RNA extracted from both serum and plasma, qRT-PCR was performed and linear recovery was obtained down to 100 lentivirus particles from just 10 uL of sample. Recovery was linear and consistent between replicates and samples.

 

ChargeSwitch® EasyPlex™ Viral RNA/DNA Kit

 
Sensitive and linear viral detection using the ChargeSwitch® EasyPlex™ Viral RNA/DNA Kit. Lentivirus (RNA virus) was inoculated into plasma and serum and 10 uL was used for processing with the ChargeSwitch® EasyPlex™ Viral RNA/DNA Kit. Amplification in the well was performed in 50 uL using the SuperScript™ III Platinum One-Step Quantitative RT-PCR System for qRT-PCR. Note that detection is sensitive and linear down to 100 viruses for both sample types, even though plasma is a more proteinaceous sample.
