The BaculoTiter™ Assay Kit rapidly determines the titer of an unknown baculovirus sample with minimal handling steps, providing both accuracy and convenience in an easy-to-use kit format. With the BaculoTiter™ Assay Kit, you can:- Obtain accurate results in two days, compared to ten-day dilution assays (Figures 1 and 2)
- Reduce inter- and intra-user variability with fewer handling steps using the ratiometric, live-cell readout (Figure 3)

How it Works
The BaculoTiter™ assay uses a stable insect cell line (Sf21) engineered to express the beta-lactamase reporter gene under control of a proprietary baculovirus-responsive promoter. Upon infection with baculovirus, BaculoTiter™ cells show a dose-dependent increase in beta-lactamase activity, which is easily quantified by a ratiometric live-cell substrate and fluorescence microplate reader with a 409/20 nm excitation filter and 460/40 nm and 530/30 nm emission filters.

Eight different virus stocks with a range of titers were compared by a limiting dilution assay and the BaculoTiter™ Assay.

Eight scientists, selected without regard to previous baculovirus experience, performed the BaculoTiter™ Assay with two previously titered virus stocks. Each user followed the assay protocol independently. The average titer obtained for each sample, the average variation from the expected titer and the inter-user variability (% CV) are indicated. The BaculoTiter™ Assay accurately determined the titer of the provided virus stocks and was highly reproducible between users.
Contents and Storage:
The BaculoTiter™ Assay Kit contains an engineered Sf21 cell line and all of the necessary detection reagents BaculoTiter™ Detection Reagents contain only the detection reagents required to determine baculovirus titers using the Sf21 cell line available in the BaculoTiter™ Assay Kit, and come packaged in larger sizes for higher-throughput titer determinations. Store as directed in the BaculoTiter™ Assay Kit and Detection Reagents protocol.
