Compare Laser Configurations

The Attune® Acoustic Focusing Cytometer comes equipped with one of the two configurations:

Red/Blue Laser Configuration
  • 638nm (50 mW) and 488 nm (20 mW) lasers
  • Four emission channels off the blue laser and 2 emission channels off the red laser
  • Up to six-color analysis in addition to forward and side scatter data collection
  • Absolute cell counts without the need for counting beads

Use this configuration with your current red/blue reagents that you are already familiar with.

Violet/Blue Laser Configuration
  • 405 nm (50 mW) and 488 nm (20 mW ) lasers
  • Three emission channels per laser
  • Up to six-color analysis in addition to forward and side scatter data collection
  • Absolute cell counts without the need for counting beads
  • Best suited for use with the Molecular Probes® violet laser-excitable reagents to achieve optimal results in many applications including:

    Cell proliferation
    • Stem cell side population
    Marine sample analysis
    Live cell cycle analysis

Choose the Laser Configuration for Your Specific Needs

The Blue/Red Configuration
Use this configuration with your current blue/red reagents that you are already familiar with. It delivers the same precision and sensitivity as the blue/violet configuration for many standard research applications. For example, the blue/red configuration shows excellent segregation of populations in immunophenotyping experiments with up to six colors (Figure 1).

Figure 1. Six-color immunophenotyping analysis on the Attune® Acoustic Focusing Cytometer with blue/red laser configuration. Mouse (C57BL/6J) peripheral blood is stained with a six-color immunophenotyping panel consisting of anti-mouse direct conjugates: (A) CD3e Alexa Fluor® 647; (B) CD19 PE-Cy®5.5; (C) CD4 PE-Cy®7, CD8 Alexa Fluor®488, CD11c APC-Cy®7, and NK1.1 PE direct antibody conjugates.

The Blue/Violet Configuration
This configuration is best suited for use with Molecular Probes® violet laser–excitable reagents to achieve optimal results in many applications, including:

In addition, the blue/violet configuration delivers optimal results using the no-lyse, no-wash protocol (Figure 2).

(A) Daughter plot of CD45+ gate showing the B cells stained with rat anti–mouse CD45R Pacific Blue™ conjugate (parameter VL1-H, violet laser, filter 450/40 nm BP) and T cells stained with hamster anti–mouse CD3 PE-Cy®5 (parameter BL3-H, blue laser, filter 530/30 nm BP). (B) A second density dot plot, a daughter of the CD3– CD45R– gate, showing the double-positive granulocytes stained with rat anti–mouse CD11b PE (parameter BL2-H, blue laser, 575/24 nm BP filter) and rat anti–mouse GR-1 Pacific Orange™ conjugate (parameter VL3-H, violet laser, 603/48 nm BP filter). The CD11b+ GR1– population represents phagocytes (monocytes, macrophages, and any circulating dendritic cells).
