Parental Cell Lines

  Gibco offers low licensing access to two established and well-characterized parental CHO host cell lines. Both the CHO DG44 and CHO-S cell lines are suspension adapted into AOF chemically defined media, banked under cGMP guidelines and are supported by complete lineage documentation packages to streamline regulatory filings.

Vector Design and Construction

   

Expression levels can be systematically enhanced by creating an optimal vector which contains the desired genetic elements, has minimal licensing requirements and royalties and is regulatory friendly. To that end, each of our CHO cell line platforms is complemented by vectors designed for high level expression. Our vectors ensure:

  • Rapid cDNA integration into a vector of your choice using Invitrogen's proprietary cloning technologies
  • Licensing agreements that ensure access to a diversity of elements with freedom to operate and clear line of sight to financial commitments through commercialization
  • A final sequence verified stable construct suitable for your expression needs

Transient Protein Expression

   

Transient transfection of mammalian cells has become increasingly important to produce milligram quantities of a recombinant protein for characterization and preclinical studies.  We can assist in:

  • Establishment of the cell line, culture conditions, expression vector, and transfection agents for the optimal transient expression system in adherent or suspension cultures
  • Use of our serum-free Freedom® expression systems to produce protein
    in suspension CHO and 293 cultures in short timeframes

Stable Cell Line Development

   

Our capabilities start with the components and proven workflows from our cell
line development kits which are then coupled with an integrated suite of
technologies that encompass:

  • Use of established serum-free media for the entire workflow leading to a
    seamless transfer into process optimization
  • Serum-free cloning to avoid animal-origin material contact and improve
    control over product development
  • High-throughput screening for the most productive clones, accomplished by ClonePix FL technology
  • Availability of integrated base and feed media platforms for clone
    screening leading to shorter process development timelines

Stability Evaluation

 

We use established industry standard protocols to identify cGMP compliant clones
by assessing :

  • Cell growth rates and proliferation
  • Protein expression stability for the product of interest

Cell Line Banking

 

We offer multiple outsource partnering options to meet the needs for GMP
preparation and characterization of Master and Working cell banks.

  • Extensive testing of any seed stock using multiple validated assays
  • GMP preparation and characterization of the Master Cell Bank and Working Cell Banks
  • Cell bank storage with strictest maintenance and security

Medium Development
