Bac-to-Bac® Baculovirus Expression System

Bac-to-Bac® The fastest method for recombinant baculovirus production
The Bac-to-Bac® Baculovirus Expression System provides the fastest method available for producing recombinant baculovirus. Use Bac-to-Bac® to express your protein of interest in as little as two weeks -- that's far faster than any other baculovirus system.
Fast protein
The Bac-to-Bac® Baculovirus Expression System is faster and easier than traditional baculovirus expression, and it maintains high levels of protein expression. Bac-to-Bac® relies on generation of recombinant baculovirus by site-specific transposition in E. coli rather than homologous recombination in insect cells.

Traditional baculovirus systems require purification and amplification of an initial low-titer viral supernatant. Bac-to-Bac®'s pFastBac™ vector, however, recombines with the parent bacmid in DH10Bac™ E. coli competent cells to form an expression bacmid. Transfect the bacmid into insect cells for fast production of a high titer of pure recombinant baculovirus particles in the very first transfection. You'll save weeks of precious time.

High expression, easy screening
pFastBac™ uses the strong polyhedrin promoter to generate high levels of expression in a variety of insect cell lines, as figure 2 demonstrates. ß-glucuronidase was expressed in Sf9, Sf21, and High Five™ cells using the Bac-to-Bac® System, with high expression levels achieved in all three cell lines.

Contents and storage

Each Bac-to-Bac® Baculovirus Expression System includes 10µ of pFastBac™1 vector, 0.5 ml Max Efficiency® DH10Bac™ chemically competent cells, pFastBac™1-Gus control vector, and 1 ml of Cellfectin® transfection reagent. The Bac-to-Bac® Vector Set contains only the expression and control vectors as described above. Store Max Efficiency® DH10Bac™ competent cells at -80°C. Store Cellfectin® at +4°C. Store the vectors at -20°C. All components are guaranteed stable for 6 months when properly stored.
