  • Product Usage: cDNAs for truncated and full length TRAF1 and TRAF2 proteins were obtained by PCR amplification using Pfu polymerase. This was followed by incubation with Taq to allow for the addition of single 3'A overhangs to enable direct direct cloning into the pCRII vector. For expression experiments, DNA fragments were cloned into pcDNA3. Expression constructs were transfected into KT-3 cells. (Note: Current version of pcDNA3 is pcDNA3.1 +/-.)

CD40 induces interleukin-6 gene transcription in dendritic cells: regulation by TRAF2, AP-1, NF-kappa B, AND CBF1.

Citations & References

  • Authors: Mann J, Oakley F, Johnson PW, Mann DA,
  • Journal: J Biol Chem (2002) 277:17125-17138
  • Product Usage: Cells were cultured in RPMI-1640 supplemented with 100 units/ml penicillin, 100 µg/ml streptomycin, 2 mM L-glutamine, and 10% fetal calf serum. Transient induction of IL-6 mRNA by anti-CD40 treatment was associated with reduced p50:p65 NF-B DNA binding and elevated binding of CBF1 to a site overlapping the NF-B site. The authors found that prolonged induction of CBF1 may contribute to the transient nature of the IL-6 response.
