For retroviral transduction, the full-length and a C-terminally truncated mutant of Smad4/DPC4 were cloned into retroviral vector and were cotransfected into 293T producer cells with retroviral packaging vectors. Retroviral particles released by 293T cells were used to infect PANC-1, and CFPAC-1 cells. Infected cells were obtained by selection with Geneticin (PANC-1, 700 µg/ml; CFPAC-1, 350 µg/ml).
