- Product Usage: Neonatal rat ventricular cardiomyocytes (NRVC cells; 0.7 x 10e6) were transfected with rat CHIP siRNA (carboxyl terminus of Hsp 70-interacting protein; 50 nmol/L) in 3 mL media with 6 uL of Lipofectamine RNAiMAX. Cells were harvested 60 hrs post-transfection. CHIP knockdown was measured via RT-PCR and westerns, and it prevented HG-induced GATA4 depletion. CHIP may be the E3 ligase that is responsible for hyperglycemia-induced GATA4 degradation.

- Product Usage: Drosophila Schneider 2 cells (S2) were grown in serum-free Sf-900 II serum-free medium at 26 °C and passaged every week. AB2.1 murine embryonic stem cells were incubated in M15 medium (Dulbecco's modified Eagle's medium, 15% fetal calf serum, 0.1 mM -mercaptoethanol, 2.0 mM glutamine, 0.05 mg/ml streptomycin, 0.03 mg/ml penicillin) 2 h prior to manipulation. ES cells were cotransfected in combinations with either pCGN (2 µg), pCGN-Nkx2-5 (2 µg), or pCGN-SRF (2 µg) by LipofectAMINE.
