Protocol and Assay Conditions
A stringent validation process ensures the highest quality data possible with each data point performed in duplicate. Strict quality control protocols ensure that any assay results not meeting set specifications will be automatically repeated, this includes a control inhibitor titration (IC50) for each kinase on every plate.
Comprehensive titrations are performed for all of our profiling kinases against Invitrogen’s panel of available substrates. In addition, ATP titrations are performed for each kinase to determine the Km[app] ATP for each target. As a result, you can choose to have your compound run at, above, or below the Km[app] ATP for each kinase in the panel. This flexibility allows you to identify specific types of inhibitors with various mechanisms of action.

2. Receive confirmation from Invitrogen with scope of project and pricing
3. Supply purchase order and prepare compounds for submission to Invitrogen
4. Receive an email report with the results within two weeks

Invitrogen is the original manufacturer of all kinases used in the SelectScreen™ Kinase Profiling Service; we ensure the consistency and reliability of each target. All kinases are of human origin; are quality controlled through mass spectrometry, SDS-PAGE, and radioactivity assays; and are the same proteins sold in the Invitrogen catalog. We will profile your compounds against any number of targets among available kinases in either single concentration characterizations (n=2) or IC50 determinations (ten points, n=2). Listed below are the kinases available for profiling, the selection is increasing frequently, so check back often for updates to this list.
