Our streptavidin, Marina Blue™ conjugate is a bright, blue-fluorescent probe with excitation/emission ~365/460 nm. Streptavidin is a nonglycosylated biotin-binding protein that is isolated from bacteria and has a near-neutral isoelectric point. It is widely used to detect biotinylated probes, because it reportedly exhibits less nonspecific binding than does the glycosylated, biotin-binding protein, avidin.

CaptAvidin™ biotin-binding protein contains a nitrated tyrosine in its biotin-binding site. This chemical modification permits dissociation of the avidin-biotin complex under milder conditions than the traditional avidin-biotin complex. At pH 4.0, CaptAvidin™ biotin-binding protein binds biotin tightly with a K a of ~10 9 M -1 . This association can then be reversed at pH 10, allowing complete dissociation of the avidin-biotin complex. This is in contrast to the harsh conditions (8 M guanidine hydrochloride, pH 1.5) needed to dissociate the avidin-biotin complex.

The pcDNA™6 BioEase™ Gateway® Biotinylation System provides an easy, efficient method for expressing, purifying, and detecting biotinylated recombinant proteins. The pcDNA™6/BioEase™-DEST vector (Figure 1) incorporates a 72 amino acid sequence from K. pneumoniae that directs in vivo biotinylation of a specific lysine residue. This K. pneumoniae tag is recognized and efficiently biotinylated in vivo by a native mammalian system similar to the enzyme Biotin Protein Ligase (BPL) in E. coli (1). Proteins produced from the pcDNA™6/BioEase™-DEST vector are expressed as fusion proteins to this sequen...

Alexa Fluor® 647 streptavidin comprises a biotin-binding protein (streptavidin) covalently attached to a fluorescent label (Alexa Fluor® dye). Streptavidin has a very high binding affinity for biotin, and a conjugate of streptavidin is commonly used together with a conjugate of biotin for specific detection of a variety of proteins, protein motifs, nucleic acids, and other molecules (for example, a biotinylated primary antibody bound to a protein target can be detected with a fluorescently labeled streptavidin). Strategies similar to this are used in many detection protocols including western ...

pHrodo™ Red Avidin is a conjugate of avidin and pHrodo™ Red dye, a novel, fluorogenic dye that dramatically increases in fluorescence as the pH of its surroundings become more acidic. pHrodo™ Red dye conjugates are non-fluorescent outside the cell, but fluoresce bright red in phagosomes, making them ideal tools for studies ranging from phagocytosis of bioparticles to receptor internalization. Original pHrodo™ and pHrodo™ Red dye are the same molecule. With pHrodo™ Red Avidin Fluorogenic pH Sensor You Get: • Specific detection of phagocytosis and endocytosis • Faster staining • More accurate re...

Biotin-14-dATP is a patented dATP analog with biotin attached at the 6-position of the purine base by a 14-atom linker. The long, 14-atom spacer arm allows accessibility to streptavidin for sensitive detection of probe-target hybrids in in situ and filter-based hybridizations. Biotin-14-dATP can be efficiently incorporated into DNA by nick translation in the presence of dCTP, dGTP, and dTTP (1). Biotin-14-dATP can be added to the 3´ end of DNA using terminal deoxynucleotidyl transferase (2). The amount supplied is sufficient for labeling up to 50 µg of DNA by nick translation. Performance and ...

The Alexa Fluor 488 Oligonucleotide Amine Labeling Kit provides a simple method for labeling amine-modified oligonucleotides with our bright, photostable, green-fluorescent Alexa Fluor 488 dye (which is spectrally similar to fluorescein). Oligonucleotides labeled with this kit can be used as primers and as probes in hybridization experiments but should not be used for automated DNA sequencing since the Alexa Fluor dye in the kit is supplied as mixed isomers. The kits have been optimized for labeling reactions with 50 µg of a 5'-amine—modified oligonucleotide, 18 to 24 bases in length.
