T669) and rhodamine 110, bis-<SPAN CLASS=\"small\">L-aspartic acid amide (Cat. no. R22122) for 15 minutes. Apoptotic cells show green cytosolic fluorescence resulting from cleavage of the rhodamine 110, bis-<SPAN CLASS=\"small\">L-aspartic acid amide substrate by active caspase-3. C2938). After a 30-minute incubation, the cells were washed and then incubated simultaneously with FM® 5-95 (Cat. no. T23360) and Hoechst 33342 (Cat. no. H1399, H3570, H21492) in phosphate-buffered saline (PBS) for an additional five minutes before washing and mounting in PBS. Actin filaments were labeled with Alexa Fluor® 488 phalloidin, and nuclei were stained with Hoechst 33342. Coverslips were mounted on slides using ProLong® Gold antifade reagent.
