The Ambion® RNA-Seq Library Construction Kit compatible with Illumina® GAII is a single kit for RNA library construction of either whole transcriptome or small RNA (such as miRNA), for use in downstream next generation sequencing analysis. Designed as a complete solution with a common workflow, the kit optimally combines all necessary reagents for discovery of coding RNA, non-coding RNA, alternative splicing, small RNAs and isoforms, while preserving strand orientation. The Ambion® RNA-Seq Library Construction Kit compatible with Illumina® GAII platform is based on the same technology used in the SOLiD™ Total RNA-Seq Kit.

Whole Transcriptome Analysis
The whole transcriptome protocol enables rapid construction of strand-specific libraries. Libraries can be constructed in just 2 days starting with as little as 100 ng of Poly(A+) RNA, 200 ng of rRNA depleted RNA, or 200 ng of total RNA.

Deeper Transcriptome Analysis
The Ambion® RNA-Seq Library Construction Kit allows for more thorough investigation of transcriptome complexity by not being limited to just Poly(A+) RNA, and the ability to create libraries compatible with paired-end sequencing. The expanded abilities of the kit allow for detection of known and unknown transcripts, including alternative splicing, fusion transcripts, and SNPs.

Small RNA Analysis
The small RNA protocol enables rapid construction of small RNA (miRNA ~10-40 nt) libraries. Detailed instructions are provided for starting with total RNA or small enriched RNA depending on the amount of small RNA contained in the sample of choice. If enrichment is required, enrichment protocols are provided.

Preserving Strand Orientation
Unlike other methods that ligate adapters to ds cDNA, the new Ambion® RNA-Seq Library Construction Kit utilizes proprietary technology to attach the adapters in a directional manner. Both the 3' and the 5' adapter are attached simultaneously, reducing ligation and clean-up steps. The resulting libraries preserve the strand information of the transcripts making up the library. This orientation of the individual strands in the libraries allows for more accurate determination of the structure and expression level of transcripts. Additionally, novel regions of transcription from both the sense and antisense genomic strands can be discovered.

Downstream Compatibility
Compatible with the Illumina® GAII sequencing platform, the Ambion® RNA-Seq Library Construction Kit contains enough reagents to process up to 12 samples for whole transcriptome or small RNA analysis.
