Description

Lipofectin® Transfection Reagent is the reagent of choice for transfectin of endothelial cells; some of our customer also use it for transfection of DNA, RNA or oligonucleotides into their cell line of choice. Lipofectin® has also been shown to work well in combination with PLUS® reagent for the transfection of HeLa cells (1).

Product Usage: To determine the optimal non-viral transfection method for human smooth muscle cells and endothelial cells, the authors transfected reporter (CAT) plasmid into these cells using seven different transfection reagents including Lipofectmine Plus, Lipofectamine 2000, Lipofectin and FuGene, two electroporationnmethods and a photochemical internalization. Electroporation is the most effective method. FuGene 6 and Lipofectamine PLUS were the preferred transfection reagents.

Product Usage: HeLa cells were maintained in Dulbecco's modified Eagle's medium (Invitrogen, Carlsbad, CA) containing 10% fetal calf serum and 0.1 mg/ml normocin-O at 37 °C in 5% CO2. Thirty-five-mm dishes of HeLa cells (70-80% confluent) were transfected with 8 µl of Lipofectin (Invitrogen) and 2 µg of pcDNA3.1/Myc-His(+), (Invitrogen) encoding the appropriate cDNA.
