Description

T-REx™ Cell Lines stably express the tetracycline repressor protein (Table 1). They save significant time and effort when using the T-REx™ System. The T-REx™ Cell Lines are functionally tested by transient transfection with the positive control vector pcDNA™4⁄TO⁄lacZ. T-REx™ Cell Lines exhibit extremely low basal expression levels in the repressed state and high expression upon induction with tetracycline (Figure 1).

Product Usage: T-Rex 293 cells were used to study the effect of down regulating VDAC1 on cell growth. The VDAC1 gene was cloned into the pcDNA4/TO vector and induced in T-REX 293 cells with 200-2500ng/ml tetracycline. Knockdown of VDAC1 was achieved by transfection of an shRNA vector by calcium phosphate transfection. Cell viability was analyzed by staining with acridine orange and ethidium bromide. Apoptosis was assayed by Annexin V-PE binding. A NADP reduction assay was used to assess ATP synthesis.

Product Usage: T-REx 293 cells were seeded in a poly-D-lysine 6-well cell culture dish 24 h prior to transfection and maintained at 37C until they were ~ 90% confluent. Cells were then transfected with 4 ug of the plasmid DNA, p-T-RExDEST30-hTf using Lipofectamine 2000. Twenty-four hours posttransfection, cells were diluted and selected in Hygromycin- B (200 ug/mL) and Geneticin(400 ug/mL). Selected clones were then expanded into cell lines and transferrin concentration was quantified by ELISA.

Product Usage: T-Rex-293 cells stably expressing pcDNA6/TR were transfected by calcium phosphate/DNA precipitation with PP2C-pcDNA4/TO. At 48 h after transfection, cells were seeded into fresh medium containing blasticidin (5 µg/ml) and zeocin (200 µg/ml). The selective medium was replaced every 3-4 days until foci became visible. Several clones were isolated and characterized. Mut-PP2C or TO stable transfected clones were pooled. Stable cell lines were maintained in medium containing blasticidin and zeocin.

After transfection, zeocin was added to the culture medium, and surviving colonies of cells were isolated and expanded into 6-well plates.
