Specific for TNKS1

TNKS1 (tankyrase, TRF1-interacting ankyrin-related ADP-ribose polymerase) is a poly-ADP-ribosyltransferase involved in various processes. Human tankyrases post-translationally modifies multiple proteins involved in telomere length maintenance, sister telomere association, spindle assembly, trafficking of glut4-containing vesicles, and insulin responsiveness. Recently tankyrases have shown to be involved in the Wnt signaling pathway, which at the heart of this pathway is the beta-catenin destruction complex. Tankyrases bind directly to axin, which is a member of the destruction complex involved in beta-catenin degradation. Inhibition of tankyrase stabilizes axin, increases the activity of the destruction complex, and promotes degradation of beta-catenin. Thus, tankyrases are an attractive target anticancer therapies.
